To systematically determine E-Cadherin protein expression in normal and cancerous tissues, 14, tumor samples from different tumor types and subtypes as well as samples of 76 different normal tissue types were analyzed by immunohistochemistry in a tissue microarray format. E-Cadherin was strongly expressed in normal epithelial cells of most organs. From 77 tumor entities derived from cell types normally positive for E-Cadherin, 35 Tumors with the highest rates of E-Cadherin loss included Merkel cell carcinoma, anaplastic thyroid carcinoma, lobular carcinoma of the breast, and sarcomatoid and small cell neuroendocrine carcinomas of the urinary bladder.

Cadherins named for "calcium-dependent adhesion" are cell adhesion molecules important in forming adherens junctions that let cells adhere to each other. Cell-cell adhesion is mediated by extracellular cadherin domains, whereas the intracellular cytoplasmic tail associates with numerous adaptors and signaling proteins, collectively referred to as the cadherin adhesome. Similar to classical cadherins, desmosomal cadherins have a single transmembrane domain, five EC repeats, and an intracellular domain. There are two types of desmosomal cadherins: desmogleins and desmocollins. These contain an intracellular anchor and cadherin like sequence ICS. The major function of desmoplakins is to bind to intermediate filament by interacting with plakoglobin, which attach to the ICS of desmogleins, desmocollins and plakophilins.
In contrast, most mutations found in e-cadherin lobular breast cancers were out-of-frame mutations, which yield secreted truncated E-cadherin products.
